A real H2 Biology application question, answered to the scheme
The problem
A drug binds to the active site of an enzyme but is not broken down by it. As the substrate concentration is steadily increased, the reaction rate of the inhibited enzyme approaches the same maximum rate (Vmax) as the uninhibited enzyme, but reaches it more slowly. Explain what type of inhibition this is and why the rate eventually recovers.
Worked solution
- 1Identify the binding site: the drug binds the active site, so it competes with the substrate for the same site β this is competitive inhibition.
- 2Explain the mechanism: while the inhibitor occupies the active site, substrate cannot bind, so at low substrate concentration the reaction rate is lowered.
- 3Explain the recovery: because the binding is reversible and the inhibitor is not consumed, raising substrate concentration increases the chance that substrate, rather than inhibitor, occupies any free active site.
- 4Reach Vmax: at sufficiently high substrate concentration the substrate out-competes the inhibitor, so essentially all active sites work on substrate and the same Vmax is reached β only more substrate is needed to get there.
Answer: Competitive (reversible) inhibition β Vmax is unchanged but a higher substrate concentration is required, so the apparent Km increases.
The marks here are won on linking observation to mechanism: 'same Vmax, reached more slowly' is the textbook fingerprint of competitive inhibition. H2 Biology rewards naming the precise term (competitive, reversible, Km) and explaining the cause, never just describing the graph.
